چکیده

Methods The various xenogeneic tissues (porcine pericardium, aortic valve, aortic wall and bovine pericardium, aortic valve, aortic wall) were fixed in 0.5% glutaraldehyde solution and implanted into the subcutaneous tissue of the wild type-mouse and the KO mouse for 3 months. The implanted mice were harvested and studied for the histopathology, amounts of calcification and immune responses by serum ELISA.

Differences in xenoreactive immune response and patterns of calcification of porcine and bovine tissues in α-Gal knock-out and wild-type mouse implantation models.

OBJECTIVES Although bioprostheses are widely used in cardiovascular surgery, their durability is limited due to degeneration. Degeneration of bioprostheses limiting its clinical use results from multiple factors, and immune reaction has been considered to be one of the most important factors. The study objectives were to compare the mechanical characteristic differences of porcine and bovine pr...

The Immune Responses and Calcification of Bioprostheses in the α1,3-Galactosyltransferase Knockout Mouse.

BACKGROUND The study aim was to evaluate the immune reaction, difference of degenerative calcification, and anti-calcification effect of decellularization with or without α-galactosidase in bovine pericardium and porcine heart valves, using an α1,3-galactosyltransferase (α-Gal) knockout (KO) mouse model. METHODS In order to elucidate the anti-calcification effect of decellularization with or ...

In vivo efficacy of alpha-galactosidase as possible promise for prolonged durability of bioprosthetic heart valve using alpha1,3-galactosyltransferase knockout mouse.

The immune response due to Galα1,3-Galβ1-4GlcNAc-R(α-Gal) epitopes is an important factor in bioprosthetic heart valve failure. The aim of this study was to evaluate the immune reaction and anticalcification effect of α-galactosidase and decellularization for glutaraldehyde (GA)/genipin fixed bovine pericardium using α1,3-galactosyltransferase knockout(α-Gal KO) mouse(C57BL/6). Bovine pericardi...

Calcification of decellularized and alpha-galactosidase-treated bovine pericardial tissue in an alpha-Gal knock-out mouse implantation model: comparison with primate pericardial tissue.

OBJECTIVES Immune reaction against the α-Gal(Galα1,3-Galβ1-4GlcNAc-R) epitope is known to be one of the important factors for calcification of bioprostheses. The genetically manipulated α-Gal knock-out (KO) mice lacking the α-Gal epitope and containing anti-α-Gal antibodies may simulate an immune response against xenoantigens in primates including humans. The aims of this study were (i) to comp...

on our article [2]. Our group proved that genipin fixation is a novel alternative to conventional glutaraldehyde fixation, and the addition of decellularization, organic solvent treatment and detoxification prevent calcification of glutaraldehyde/genipin-fixed bovine pericardium. We also found that the titer of anti-α-Gal(Galα1-3Galβ1-4GlcNAc-R) antibodies (immunoglobu-lin G) increases after th...
